Pretreatment with 5’-N-ethylcarboxamidoadenosine provides partial improvement in intestinal ischemia- reperfusion injury of rat

Amaç: Adenozin ve adenozin A1 reseptör agonistleri, çeşitli dokuların reperfüzyon harabiyetine karşı önkoşullanma yoluyla koruyucu etkiler göstermektedir. Çalışmanın amacı, sıçan ince bağırsağının reperfüzyon harabiyeti üzerine adenozin A1/A2 reseptör aktivasyonunun etkilerini incelemekti. Yöntemler: Denekler herbiri rastgele sekiz hayvan içeren dört gruba ayrıldı: sham kontrol, iskemi-reperfüzyon kontrol, 5’-N-etilkarboksiamidoadenozin (NECA) (non-selektif A1 /A2 agonisti, 0.1 mg/kg, i.v.) tedavili iskemi-reperfüzyon ve teofilin (non-selektif A1 /A2 antagonisti, 20 mg/kg, i.v.) tedavili iskemi-reperfüzyon. Tedaviler iskemi yapılmad . . .an 5 dk önce uygulandı. Süperiyor mezenter arter 30 dk klempe edildikten sonra reperfüzyon dönemi 180 dk sürdü. Terminal ileum segmentlerinin, KCl, karbakol ve substans P’ye olan kasılma yanıtlarının kaydedilmesinin yanısıra, dokuların miyeloperoksidaz, malondialdehit ve indirgenmiş glutatyon miktarları da ölçüldü. Bulgular: İskemi, nötrofil infiltrasyonu ve lipid peroksidasyonunu ileri düzeyde yükseltirken aynı zamanda indirgenmiş glutatyonu düşürdü. Sham control grubuy- la karşılaştırıldığında, kasılma yanıtları iskemi-reperfüzyon grubunda ciddi düzeyde azaldı. NECA ön tedavisi, doku indirgenmiş glutatyon miktarını ileri derecede düzeltti ve ayrıca iskemi-reperfüzyon sonucu azalmış kasılma yanıtını kısmen iyileştirdi. Teofilin ile ön tedavi ise herhangi koruyucu bir etki göstermedi. Sonuç: Adenozin A1 /A2 reseptör aktivasyonunun, iskemi sonucu gelişen bağırsak kasılma bozukluğuna karşı kısmi koruma sağladığını ve ayrıca bu korumanın, indirgenmiş glutatyon düzeyinin fizyolojik seviyede tutulması yolu ile muhtemelen gerçekleştiğine dair ek kanıt önermekteyiz. Objectives: Adenosine and adenosine A1 receptor agonists exert protective effects against reperfusion injury in different tissues by mediating preconditioning. The aim of the present study was to examine the effects of adenosine A1 /A2 receptor activation on reperfusion-induced small intestinal injury in rat. Methods: Animals were randomized into four groups each including eight as following: sham control, ischemia-reperfusion control, 5’-N-ethylcarboxamidoadenosine (NECA) (non-selective A1 /A2 agonist, 0.1 mg/kg, i.v.)-treated I/R, and theophylline (non-selective A1 /A2 antagonist, 20 mg/kg, i.v.)-treated I/R groups. The treatments were administered 5 min before inducing ischemia in which superior mesenteric artery was clamped for 30 min followed by 180 min of reperfusion pe- riod. Myeloperoxidase, malondialdehyde, and reduced glutathione contents of terminal ileum samples were measured besides recording contractile responses to KCl, carbachol and substance P. Results: Ischemic insult significantly increased neutrophil infiltration and lipid peroxidation while decreasing the reduced glutathione. Contractile responses were seriously reduced in I/R group compared to that of the sham control group. NECA pretreatment alleviated the tissue content of reduced glutathione remarkably besides providing partial amelioration of I/R-reduced contractile response, while theophylline pretreatment had no any protective effect. Effects of ozonated olive oil on acute radiation proctitis in rats

Background: Acute radiation proctitis is a common complication of pelvic radiation and management of acute radiation proctitis is under evaluation. The beneficial effects of ozonated olive oil (OzOO) have already been shown in the treatment of chronic wounds. Thus, this study was designed to evaluate the therapeutic effects of topical OzOO on acute radiation proctitis. Aims: To evaluate the therapeutic effects of topical OzOO on acute radiation proctitis. Study Design: Animal experimentation. Methods: Rats were divided into three groups: control; irradiation+saline (1 mL); and irradiation +OzOO (1 mL). A single fraction of 17.5 Gy w . . .as delivered to each rat. The OzOO was administered rectally each day after irradiation. Each rat was observed daily for signs of proctitis. Irradiated rats were euthanised on days 5 and 10. The mucosal changes were evaluated macroscopically and pathologically. Results: According to the clinical findings, five rats in the irradiation+saline group showed Grade 4 symptoms on the 10th day. Macroscopic finding scores on the 10th day in the irradiation+saline and irradiation+OzOO groups were statistically significantly different. On pathological examination, radiationinduced mucosal damage was the most prominent 10 days after irradiation in saline-treated rats. On the 10th day, the irradiation+OzOO group showed mild inflammation and slight crypt change, which corresponded to Grade 1 pathological findings. Curcumin reduces oxidative stress in ovariectomized rat brain subjected to chronic cerebral hypoperfusion

Giriş: Turmeriğin majör komponenti olan kurkumin hem antioksidan hem de antiinflamatuar aktivite göstermektedir. Çalışmamızda, kronik serebral hipoperfüzyon modelini kullanarak, overleri alınmış dişi sıçan beyininde, kurkuminin oksidatif stresi azaltıp azaltmadığını araştırdık. Gereç ve Yöntem: Kronik serebral hipoperfüzyon, her iki karotid komunis arterin kalıcı ligasyonu ile sağlandı. Denekler (4-6 aylık toplam 30 adet yetişkin Wistar Albino dişi sıçanlar) rastgele üç gruba ayrıldı: sham kontrol, iskemi ve 14 gün boyunca günlük kurkumin (100 mg/kg) verilen iskemi grubu. Ligasyon sonrası 14üncü günde tüm gruplardaki beyin dokuların . . .ın malondialdehid (MDA) ve indirgenmiş glutatyon (GSH) içerikleri ölçüldü. Bulgular: İskemi, MDA içeriğinin ileri dercede yükselmesine neden olurken GSH seviyesini anlamlı derecede azalttı. Diğer yandan, kurkumin tedavisi iskemik beyin dokusunda anlamlı derecelerde MDA düzeyini düşürerek ve GSH içeriğini yükselterek, değerlerin sham grubundakiler seviyesine geri dönmesini sağladı. Sonuç: Elde ettiğimiz sonuçlar, vasküler demansın deneysel hayvan modeli olan kronik serebral hipoperfüzyonda kurkuminin hem oksidatif stresi hem de lipid peroksidasyonunu azalttığını önermektedir. Altta yatan moleküler mekanizmaların derinlemesine daha fazla araştırılması sonrasında inanıyoruz ki kurkuminin tedavideki verimliliği, klinik uygulamalar için özellikle de vasküler demanslı menopoz sonrası yaşlı kadınlarda test edilmesine layık olacaktır. Introduction: Curcumin, the major constituent of turmeric, exhibits both antioxidant and anti-inflammatory activities. In the present study, we investigated whether or not curcumin reduces oxidative stress in ovariectomized female rat brain by using a model of chronic cerebral hypoperfusion. Materials and Method: Chronic cerebral hypoperfusion was induced by permanent ligation of both common carotid arteries. Animals (a total of 30 adult female Wistar Albino rats, 4-6 months old) were randomly divided into three groups: sham control, ischemia, and ischemia plus daily curcumin treatment (100 mg/kg) for 14 days. At day 14 after the ligation, malondialdehyde (MDA) and reduced glutathione (GSH) contents of brain tissues were measured in all groups. Results: Ischemia caused a significant increase in MDA content but a meaningful decrease in GSH levels. Treatment with curcumin, however, lowered MDA and elevated GSH contents significantly in ischemic brain tissue, bringing their levels back to that of the sham group. Conclusion: Our results suggest that curcumin attenuates both oxidative stress and lipid peroxidation in chronic cerebral hypoperfusion, which is an animal model of vascular dementia. Redox polymerization of N-isopropylacrylamide by using hydroxylated soya oil polymer

Abstract: Water soluble hydroxylated soya oil polymer was used in the redox polymerization of N-isopropylacrylamide (NIPAM) in order to obtain water-based hydroxylated-soya oil polymer-g-PNIPAM graft copolymer. For this purpose, soya oil was exposed to air under sunlight to obtain autoxidized soya oil polymer (PSy-ox). PSy-ox was allowed to react with diethanol amine to obtain hydroxylated soya oil polymer (hydroxylated-PSy). An ammonium persulfate with hydroxylated-PSy redox initiating system was used in the polymerization of NIPAM in order to obtain thermo-responsive hydroxylated-PSy-g-PNIPAM water-based graft copolymers. Partial purification and characterization of an extracellular metallopeptidase produced by bacillus amyloliquefaciens FE-K1

The aim of this study was to purify and characterize the peptidase of Bacillus amyloliquefaciens (Fukumoto) (strain FE-K1) isolated from ropey bread. Peptidases were purified from crude enzyme solution by affinity chromatography with an efficiency of 25 % and a purification coefficient of 1.53. The optimum pH of partially purified peptidase (PPPase) solution was determined as 7.5 and the peptidases retained approximately 90 % of their initial activity in the pH range 7.0-8.5 following incubation at 37°C for 2 h. The optimum temperature for the PPPase was 60°C. The approximate molecular weight of the PPPase was determined as 36 kDa. . . .Inactivation of the PPPase in the presence of O-FEN and EDTA showed them to be metallopeptidases and 5 mM of K+1 and 5 mM of Mn+2 ions increased the enzyme activity by 4 % and 6.15 %, respectively. Comparison of the efficiencies of buffers containing ankaferd and chitosan on hemostasis in an experimental rat model with femoral artery bleeding

Objective: In the first assessment of trauma patients with major vascular injuries, we need effective and rapid-acting homeostatic materials. In this study we compare the efficiencies of Ankaferd Blood Stopper (R) and a chitosan linear polymer (Celox (R)) in an experimental rat model with femoral artery bleeding. Materials and Methods: Thirty male Wistar albino rats weighing 200250 g were divided into 3 groups: control, Ankaferd, and chitosan. The femoral artery and vein were visualized and bleeding was started by an incision. The bleeding time was recorded and categorized as 'bleeding stopped at the second minute', 'bleeding stoppe . . .d at the fourth minute', and 'unsuccessful' if bleeding continued after the fourth minute. Results: In the control group, 60% of the bleeding did not stop. In the first 4 min in the Ankaferd group, the bleeding stopped in all rats; only in 1 of the rats in the chitosan group did the bleeding not stop. In stopping the bleeding in the first 4 min, Ankaferd was similar to chitosan but better than the control group; the chitosan group was similar to the control, but the p-value was close to significance. Conclusion: For major arterial bleeding, the main treatment is surgical bleeding control, but outside of the hospital we can use buffers containing Ankaferd and chitosan on the bleeding region. The results of this study should be supported with larger studies. Furthermore, in our study, healthy rats were used. The relation between gleason score, and nuclear size and shape factors in prostatic adenocarcinoma

Aim: The aim of this study was to assess the size and shape characteristics of prostatic adenocarcinoma cell nuclei using a computer-assisted analysis system, and to compare the results with the Gleason score. Materials and Methods: Morphometric nuclear parameters, such as roundness factor, form ellipse, area, length, and perimeter, were evaluated based on specimen slides of 130 prostatic adenocarcinoma cases (77% needle biopsies and 23% prostatectomy specimens) using a computerized image analysis system. Correlation analysis between Gleason score and morphometric results was performed. Results: The Gleason score was correlated with . . . mean nuclear area (r = 0.516, P = 0.01), mean nuclear length (r = 0.298, P = 0.01), and mean nuclear perimeter (r = 0.303, P = 0.01) for all specimens. In the needle biopsy group the Gleason score was correlated with mean nuclear area (r = 0.522, P = 0.01), mean nuclear length (r = 0.398, P = 0.01), and mean nuclear perimeter (r = 0.432, P = 0.01), whereas in the prostatectomy group the Gleason score was correlated only with mean nuclear area (? = 0.619, P = 0.01) and mean nuclear roundness factor (? = -0.425, P = 0.05). Conclusions: Nuclear size and shape factors, especially mean nuclear area, were concordant with the Gleason score. Nonlinear acoustic echo cancellation using an adaptive Hammerstein block structure based on a generalized basis function

We investigated adaptive algorithms for a Hammerstein block structure in which a static nonlinear block and dynamic linear block are cascaded. The approach considered here is to use generalized orthonormal basis functions in a Hammerstein block structure by using xed pole lter banks. We applied the normalized least mean square approach to the developed adaptive algorithm in order to acquire Hammerstein block structure parameters. Performance comparison of the proposed approach was investigated considering convergence speed and parametric complexity for acoustic echo cancellation application.
