Laboratory assays of the effects of oxfendazole on biological parameters of Galleria mellonella (Lepidoptera: Pyralidae)

Galleria mellonella L. larvae were reared on a standard diet amended with varying concentrations (0, 0.0015, 0.015, 0.15, and 1.5%) of the benzimidazole-derivative antihelmintic drug oxfendazole. Survivorship, developmental times, longevity, fecundity, and fertility were monitored over the treatments. Relative to the untreated control, exposure to diet containing 1.5% oxfendazole significantly decreased survivorship in larval, pupal, and adult stages, prolonged the time to reach the adult stage, and reduced adult longevity. Glycoalkaloids are secondary metabolites commonly found in Solanaceae plants. They have anti-bacterial, anti-fungal and insecticidal activities. In the present study we examine the effects of potato and tomato leaf extracts and their main components, the glycoalkaloids ?-solanine, ?-chaconine and ?-tomatine, on development and reproduction of Drosophila melanogaster wild-type flies at different stages. Parental generation was exposed to five different concentrations of tested substances. The effects were examined also on the next, non-exposed generation. In the first (exposed) generation, addition of each extract reduced the number . . .of organisms reaching the pupal and imaginal stages. Parent insects exposed to extracts and metabolites individually applied showed faster development. However, the effect was weaker in case of single metabolites than in case of exposure to extracts. An increase of developmental rate was also observed in the next, non-exposed generation. The imagoes of both generations exposed to extracts and pure metabolites showed some anomalies in body size and malformations, such as deformed wings and abdomens, smaller black abdominal zone. Electrochemical, continuous-flow determination of p-benzoquinone on a gold nanoparticles poly(propylene-co-imidazole) modified gold electrode

A novel continuous flow biosensor based on gold nanoparticles and poly(propylene-co-imidazole) was developed for the online determination of p-benzoquinone. The amperometric response was measured as a function of p-benzoquinone concentration at an applied potential of -50 mV. The hydrogen peroxide concentration was optimized and fixed at 1 mM in samples. The mass transfer resistance of the copolymer film was minimized, and the flow cell was regenerated quickly at 1 mL/min. The resulting device provided good analytical performance based on a linear dynamic range from 5–100 µM, a short response time of 3 s, a detection limit of 3.3 µM . . ., excellent repeatability with a relative standard deviation of 0.82%, long-term stability of 95% after four weeks, and an accuracy of 105%. The gold nanoparticles enhanced the electron transfer rate on the electrode. Only a small percentage of insect species are pests. However, pest species cause significant losses in agricultural and forest crops, and many are vectors of diseases. Currently, many scientists are focused on developing new tools to control insect populations, including secondary plant metabolites, e.g., alkaloids, glycoalkaloids, terpenoids, organic acids and alcohols, which show promise for use in plant protection. These compounds can affect insects at all levels of biological organization, but their action generally disturbs cellular and physiological processes, e.g., by altering redox balance, hormonal regulation, neuronal sign . . .alization or reproduction in exposed individuals. Secondary plant metabolites cause toxic effects that can be observed at both lethal and sublethal levels, but the most important effect is repellence. Plants from the Solanaceae family, which contains numerous economically and ecologically important species, produce various substances that affect insects belonging to most orders, particularly herbivorous insects and other pests. Many compounds possess insecticidal properties, but they are also classified as molluscides, acaricides, nematocides, fungicides and bactericides. In this paper, we present data on the sublethal and lethal toxicity caused by pure metabolites and crude extracts obtained from Solanaceae plants. Büyük bal mumu güvesi Galleria mellonella L. mikrobiyal enfeksiyonların ve klinik ilaçların çalışılmasında memeli modellerine alternatif bir model olarak bilindiği gibi insektisit etkinliği çalışmalarında da model böcek olarak yaygın kullanıma sahiptir. Bu böceğin larvaları yapay besin ortamında beslenerek fluorokinolon sınıfından bir antibiyotik olan gemifloksasinin böceğin eşey oranı, dişi ve erkek ömür uzunluğu, yumurta verimi, açılma oranı gibi ergin biyolojik özellikleri üzerine etkisi laboratuvar şartlarında incelendi. Böceğin birinci evre larvaları %0.001, 0.01, 0.1 ve 1.0 oranında gemifloksasin içeren yapay besinler ile ergi . . .n evreye kadar beslendi. Gemifloksasinin denenen konsantrasyonlarını içeren besinler erkek ve dişi eşey oranı ile erginlerin ömür uzunluğu üzerinde etkili olmadığı halde yumurta verimini önemli derecede düşürdüğü belirlendi. Artan besinsel gemifloksasin konsantrasyonları ile ters orantılı olarak yumurta verimi ve açılma oranı azaldı. Kontrol besini ile yetiştirilen dişiler günde 134.46 adet yumurta üretirken gemifloksasinin yüksek miktarlarında dişiler 26.75 ve 53.5 yumurta bıraktılar. Yumurtaların açılma oranı da gemifloksasinin %0.01'lik konsantrasyonundan itibaren önemli derecede azalmış olup, en yüksek konsantrasyon bu oranı %53.71'e düşürdü. Bu çalışma erginlerin biyolojik özelliklerinin gemifloksasin tarafından önemli derecede etkilendiğini ve bu etkilerin antibiyotiğin konsantrasyonlarına bağımlı değişimler olduğunu gösterdi. Greater wax moth, Galleria mellonella L. is most commonly used as model insects for studying insecticidal efficiency of chemicals as well as it has been known as an alternative model to mamalian model for studying microbial infections and clinical drugs. The effects of a fluoroquinolone antibacterial agent, gemifloxacin, on sex ratio, male and female adult longevity, fecundity and hatchability of this insect were investigated by rearing the first instar larvae on artificial diets in the laboratory condition. The insect was reared from first-instar larvae to adult stage on an artificial diets containing gemifloxacin at 0.001, 0.01, 0.1 or 1.0%. Gemifloxacin did not affect male and female sex ratio and adult longevity while it significantly decreased egg number. Fecundity and hatchability were significantly and inversely decreased by increasing gemifloxacin concentrations. The females reared from control diet produced 134.46 per day, whereas high concentrations of gemifloxacin decreased the egg number to 26.75 and 53.5. Hatchability was also significantly decreased by 0.01% and above concentrations of gemifloxacin whereas, the highest concentration of antibiotic lowered the hatchability to 53.71%. Amphiphilic poly(3-hydroxylalkanoate) (PHA) copolymers find interesting applications in drug delivery. The aim of this study was to prepare nucleic acid adsorbed on (PHB-b-PEG-NH2) nanoparticle platform for gene delivery. For this purpose, PHB-b-PEG-NH2 block copolymers were synthesized via transesterification reactions. Silver nanoparticle incorporation effect on mechanical and thermal properties of denture base acrylic resins

Objective: The aim of the present study was to evaluate the mechanical and thermal characteristics of two denture base acrylic resins containing silver nanoparticles (AgNPs). Material and Methods: Two different acrylic denture base resins (heat-polymerized and microwave polymerized) containing 0.3, 0.8 and 1.6 wt% AgNPs were evaluated for flexural strength, elastic modulus and impact strength. The glass transition temperature (Tg) and relative heat capacity (Cp) of the samples were determined from the Differential Scanning Calorimetry (DSC) results. For statistical analysis, two-way ANOVA and Tukey-HSD tests were performed. Results: . . . Addition of 0.8% and 1.6% AgNPs in microwave-polymerized resin significantly decreased the transverse strength and elastic modulus. In terms of impact strength, the addition of AgNPs has no effect on both resin groups. Glass transition temperature (Tg) was decreased with the addition of AgNPs for both denture base resins. Conclusions: The incorporation of AgNPs, generally used for antimicrobial efficiency, affected the transverse strength of the denture base acrylic resins depending on the concentration of nanoparticles. Design of a mediated enzymatic fuel cell to generate power from renewable fuel sources

The present work reported a compartment-less enzymatic fuel cell (EFC) based on newly synthesized Poly(pyrrole-2-carboxylic acid-co-3-thiophene acetic acid) film containing glucose oxidase and laccase effectively wired by p-benzoquinone incorporated into the copolymer structure. The resulting system generated a power density of 18.8 µW/cm2 with 30 mM of glucose addition at +0.94 V at room temperature. Improvements to maximize the power output were ensured with step-by-step optimization of electrode fabrication design and operational parameters for operating the system with renewable fuel sources. We demonstrated that the improved fu . . .el cell could easily harvest glucose produced during photosynthesis to produce electrical energy in a simple, renewable and sustainable way by generating a power density of 10 nW/cm2 in the plant leaf within 2 min. A series of novel N-and N,N'-bis(quinonyl)amines were synthesized from the reactions of 2,3-dichloro-1,4-naphthoquinone (1) and polyoxypropylenediamines. The novel quinone-amine polymers were treated with some thiols in next step and N,S-substituted compounds were obtained. The structures of the novel products were characterized by micro-analysis, 1H NMR, UV-VIS, 13C NMR, and MS. The electrochemical properties of some of the novel amine-quinone polymers derivatives were also investigated by cyclic voltammetry.
